Overview

Seizures are a $ 10 billion market, served primarily by symptom based treatment approaches that focus on inhibiting nerve function.

The issued patent identified the underlying pathogenesis causing the hypersensitization of nerves and abnormal muscle contractility in most seizures. The patent’s approach focuses on inhibiting the underlying cause of the disease, rather than trying to manage its resulting symptoms.

More specifically, the patent validated that known seizure and migraine triggers mediate their actions by enhancing release of Ca++ from bone into the extracellular fluid. This in turn depolarizes nerve membranes (i.e. lowers the threshold potential for firing), increases calcium channel mediated neurotransmitter release, and increases muscle contractility by removing of the tropomyosin block between actin and myosin.

The specific claim of the patent is for use of the bone homeostasis drug raloxifene to inhibit the underlying pathogenesis. Raloxifene mimics the action of estrogen in bone homeostasis pathways, providing triple action protection against release of Ca++ into the extracellular fluid (i.e. by suppressing RANKL production, increasing output of OPG, and promoting osteoclast apoptosis).

When used concurrently, the approach would also solve the bone loss issue associated with some of today’s treatment approaches.

Raloxifene came off composition of matter patent protection in 2012 and this patent would provide patent protection for its use to treat seizures until the year 2030.

Because this is a repurposing, trials can be started in Phase II setting, at low risk, low cost, and a fast exit or implementation.
